Design Notes

This composite part was synthesized as a whole unit by IDT. Therefore there are no Biobrick scars. Instead a spacer "TCATAGAG" was included following R0011 and "TACTAGATG" following J61101 to improve translation. There is an inducible lac promoter included for expression of the LTNF10 protein. The LTNF protein sequence has a BamHI restriction nuclease site in the very middle of it. This can be used for the purpose of diagnostic digest. Both a Myc and 6xHis tag are added on towards the end of the part. These tags are used to help purify and identify this protein after expression. The Human Rhino Virus site has been added as a cleavage point to remove the two tags after purification of the protein.
